Job details

Achilles is a fast paced, trail blazing company focused on the development of the next generation of personalised T cell therapies to treat cancer. We are breaking new ground with our science - there is no playbook to copy. This means we must innovate, adapt and collaborate across our organisation, working together to solve novel problems.

We use DNA sequencing data from each patient, together with our proprietary bioinformatics platform, to identify clonal neoantigens specific to that patient, enabling the development of truly personalised T cell therapies. Originating from pioneering research at UCL, CRUK and the Francis Crick Institute, we are using our ground-breaking immuno-oncology approach to rapidly advance therapies.

Role Purpose

Single cell RNA sequencing is a cutting-edge technology that enables the detection and quantification of gene expression in individual cells, revealing the diversity and function of cells in their native environment. It is providing information at unprecedented levels of detail in many fields of science and is also a powerful tool for cell therapy development as it can help identify new cell types, states, and interactions.

As Senior Scientist, Single Cell Sequencing at Achilles Therapeutics, you will be part of the Translational Science department, more specifically you will be an integral member of the Product Phenotyping team and will support the team's efforts to gain a deeper understanding of different T cell populations within the tumour and Achilles' cell therapy products.

In this role you will generate single cell RNAseq and TCRseq data and optimise the required assays as necessary. You will plan and perform complex laboratory experiments and drive the generation of single cell sequencing data within the company. You will also work closely with members of the Translational Science Immune Monitoring and Process Development teams to carry out and organise assays on human samples from our clinical trials and research studies as well as with our Translational Analysis team to analyse these datasets.

  • The primary responsibility of the role holder will be to independently run Achilles' single cell capture and processing workflow to generate single cell RNAseq and TCRseq data
  • In addition the role holder will be expected to contribute to the wider Translational Science projects at Achilles by:
  • Performing Illumina sequencing of samples
  • Performing RNA extractions
  • Performing multicolour flow cytometry experiments
  • Processing blood samples from clinical trials
  • Designing and implementing assay development and improvements
  • Following established SOPs

Essential Experience:

  • PhD in immunology, cell biology or related field or equivalent length and depth of experience preferably in the pharmaceutical or biotechnology industry
  • Practical experience in molecular biology and genomics applications, particularly those pertaining to single cell RNA sequencing
  • Practical experience with immunoassays, including multi-parameter flow cytometry
  • Ability to test, implement and troubleshoot new approaches and techniques
  • Writing and reviewing of SOPs
  • Practical experience in human primary cell culture
  • Excellent planning, organisational and time management skills, and attention to detail
  • Team Player: Ability to learn from senior and junior members of the team
  • Good communication and interpersonal skills
  • Self-motivated with strong initiative and drive
  • Proficiency with MS Office

Desirable experience:

  • Practical experience in fluorescently activated cell sorting (FACS)
  • Practical experience in high-throughput sequencing on Illumina sequencers
  • Experience working in a GCLP (or GxP) laboratory
  • Writing and reviewing SOPs to GCLP standards

Achilles Therapeutics’ mission is to develop next-generation, patient-specific therapies that harness the immune system to destroy cancer cells. The therapies will target truncal (also known as clonal) tumour neo-antigens, unique flags to the immune system present on the surface of every cancer cell. These truncal neo-antigens are created early in the cancer’s evolution and present on all cancer cells in an individual patient’s tumour but not on healthy cells, so could allow scientists to target and destroy tumours without harming healthy tissues. Achilles Therapeutics brings together four scientific Founders with world leading expertise in the understanding of cancer evolution, bioinformatics and the development of immunotherapies.
